How Should I ‘properly’ Respond To A Job Application Which Requires Relocation?

I applied for a job in a different part of the state, knowing that I would have to relocate to that area. I want to know what would be the best way to respond (via email) that I have considered the ‘relocation’ and am still interested in the job, depending on salary and benefits (this not mentioned at this point of course). Should I ask about a relocation benefit? During interview or during salary negotiations?
