How To Put A Fine Grid Over A Picture, Which Software?

I have a picture with various sized squares. I want to put a fine grid (the size of each smallest square will be very small). Whose dimensions are known to me. I want to find how many smallest squares are occupied by each square in the picture and find their areas. Can anybody of you suggest a clear way or a software that allows me to do this?
